The explosion occurred at a police headquarters in the city of Gori, some 80 kilometers west of Georgia's capital Tbilisi.
Interior Ministry spokesman Guran Gonadze said the blast nearly destroyed the building.
Deputy Prime Minister Giorgi Baramidze says the government will deal with the perpetrators in a "very serious" manner.
"The police are already on the scene and intensive work is under way. In whatever event, appropriate action will be taken. No stone will be left unturned. I have no doubt that we will establish who did this and who gave the order. All those who were involved in this crime will be punished," Baramidze said.
Georgian Defense Minister Irakly Okruashvili says authorities are treating the blast as a terrorist attack.
There has been no immediate claim of responsibility for the blast.
